3’, 4’-Dihydroxyflavone enhances all-<i>trans</i> retinoic acid-induced superoxide-generating activity through up-regulating transcription of gp91-phox in human monoblastic U937 cells, as opposed to flavone and other hydroxyflavone derivatives

Flavones are belonging to flavonoids group and show diverse biological functions. Therefore, they have much attention as drugs for maintaining human health via contributing prevention treatment of various diseases like cancers, diabetes, neurodegenerative diseases, ischemic stroke, inflammation cardiovascular diseases. On the other hand, monoblastic leukemia U937 cells been used an excellent in vitro model system macrophage development induced response reagents such all-trans retinoic acid (RA). Here, we investigated effects flavones (flavone its hydroxy derivatives) on RA-induced O2--generating activity cells. Very interestingly, at a concentration 20 μM, 3’, 4’-dihydroxyflavone caused up-regulation (to ~ 170%) although flavone hydroxyflavone derivatives tested showed remarkable inhibitory activity. The promoting maximum value 10 μM. Semiquantitative RT-PCR immunoblotting revealed that μM up-regulates enhancing gene expression gp91-phox (mRNA level: 160%, protein 200%) while 5, 7-dihydroxyflavone 4’, 7-tetrahydroxyflavone down-regulate inhibiting p47-phox. These findings also there may be risks involved use phytochemical mixtures.
